- 요약The MEI5 gene of Saccharomyces cerevisiae is meiosis-specific and is expressed during mid/late meiotic Prophase I. A mei5$\Delta$ null mutant strain tightly arrests at the pachytene stage in meiosis. mei5$\Delta$ mutant cells arrest with highly condensed and paired homologs. Wild type appearing synaptonemal complexes are assembled and the spindle pole bodies are duplicated but unseparated during meiotic arrest of the mei5$\Delta$ mutant strain. The mei5$\Delta$ arrest is alleviated by a spo11$\Delta$ mutation and is not rescued by a spo13-1 mutation, suggesting that a MEI5 function is required after SPO11 and before SPO13 activity.
- 초록/해제
- 요약The kinetics and level of induction of non-reciprocal recombination events are modestly reduced or unaffected after induction of meiosis and return to vegetative growth of mei5$\Delta$ mutant cells. In contrast, mei5$\Delta$ mutant cells exhibit severely reduced levels of reciprocal recombination in these experiments.
- 초록/해제
- 요약During meiosis of a mei5$\Delta$ mutant strain, DNA double-strength breaks are formed on schedule, similar to that observed in a wild type strain but they persist. Heteroduplex DNA recombination intermediates are formed during meiotic Prophase I in a mei5$\Delta$ mutant strain but mature reciprocal recombination products are not detected. The RAD51 gene, when present on a high-copy number plasmid, partially suppresses the mei5$\Delta$ mutant phenotype.
- 초록/해제
- 요약The MEI5 gene product is immuno-localized to synapsed bivalents with a punctate staining pattern, as visualized with anti-Mei5 antisera.
